Production of Rhodotorula glutinis: a yeast that secretes alpha-L-arabinofuranosidase Electron. J. Biotechnol.
Martínez,Claudio; Gertosio,Cecilia; Labbe,Angelica; Pérez,Rafael; Ganga,Maria Angélica.
Rhodotorula glutinis is a yeast that secretes the enzyme alpha-L-arabinofuranosidase (E.C. 3.2.1.55) into the culture medium and thus has an interesting biotechnological potential. To determine improved culture conditions of this organism, different factors of the culture media were evaluated such as the use of peptone as nitrogen source, salts composition, pH and growth temperature. Likewise, beet molasses and beet cosette were tested as industrial carbon sources to induce the production of the enzyme and how they influence the yeast growth. Based on these studies a culture medium is proposed for growth of this yeast in a continuous system. By assaying different dilution rates an average specific activity for the enzyme of 82.4 U/mg of protein was obtained.

Proteome of fraction from Tityus serrulatus venom reveals new enzymes and toxins J. Venom. Anim. Toxins incl. Trop. Dis.
Amorim,Fernanda Gobbi; Longhim,Heloisa Tavoni; Cologna,Camila Takeno; Degueldre,Michel; Pauw,Edwin De; Quinton,Loïc; Arantes,Eliane Candiani.
Abstract Background: Tityus serrulatus venom (Ts venom) is a complex mixture of several compounds with biotechnological and therapeutical potentials, which highlights the importance of the identification and characterization of these components. Although a considerable number of studies have been dedicated to the characterization of this complex cocktail, there is still a limitation of knowledge concerning its venom composition. Most of Ts venom studies aim to isolate and characterize their neurotoxins, which are small, basic proteins and are eluted with high buffer concentrations on cation exchange chromatography. The first and largest fraction from carboxymethyl cellulose-52 (CMC-52) chromatography of Ts venom, named fraction I (Fr I), is a mixture of...

Pseudomonas spp. and P. fluorescens: population in refrigerated raw milk Ciência Rural
Almeida,Kelly Molin de; Bruzaroski,Samera Rafaela; Zanol,Daniel; Melo,Marcela de; Santos,Joice Sifuentes dos; Alegro,Lina Casale Aragon; Botaro,Bruno Garcia; Santana,Elsa Helena Walter de.
ABSTRACT: Raw milk samples were collected from cooling tanks (after they cooled for 48 h) in five dairy farms and the corresponding bulk tank (bulk milk transportation, BMT) when they arrived to the industry. Routine physical chemical analyzes and quantification of psychrotrophic ( Pseudomonas spp. and P. fluorescens ) and aerobic mesophilic (AM) populations were performed. Only relative density and titratable acidity values for samples of milk from three farms were in agreement to the quality parameters required by law. In the BMT, only the protein content has not reached the minimum value established by law, and counting was performed for AM (>105 colony forming units (CFU) mL-1) and psychrotrophic bacteria (2.8x106CFU mL-1). Pseudomonas spp....

Relations between bacterial extracellular enzyme activities and heterotrophic substrate uptake in a brackishwater environment. ArchiMer
Hoppe, H.
Extracellular enzymes mediate the decomposition of polymeric organic compounds in natural waters. In many cases these enzymes comprise a component linked to the bacterial fraction of the aquatic community and react in close association with these cells. An annual survey of microbial activities in a brackish water fjord exhibited an excellent correlation between V sub(m) of proteases (ability to split the substrate analogue methylumbelliferyl-leucine) and V sub(m) as well as T sub(R) for leucine uptake by microorganisms. Hydrolysis rates (H sub(R)) for the decomposition of naturally occurring competitive substrate analogues of the model substrates (methylumbelliferyl-(MUF)-leucine, MUF- alpha -D-glucose, MUF-N-acetyl-glucosamine, MUF-phosphate) drastically...

ROLE OF ENZYMES IN THE REMEDIATION OF POLLUTED ENVIRONMENTS J. Soil Sci. Plant Nutr.
Rao,M.A; Scelza,R; Scotti,R; Gianfreda,L.
Environmental pollution is growing more and more due to the indiscriminate and frequently deliberate release of hazardous, harmful substances. Research efforts have been devoted to develop new, low-cost, low-technology, eco-friendly treatments capable of reducing and even eliminating pollution in the atmosphere, the hydrosphere and soil environments. Among biological agents, enzymes have a great potentiality to effectively transform and detoxify polluting substances because they have been recognized to be able to transform pollutants at a detectable rate and are potentially suitable to restore polluted environments. This brief review will examine some classes of pollutants and enzymes capable of transforming them effectively into innocuous products....

Temperature-dependent alterations in metabolic enzymes and proteins of three ecophysiologically different species of earthworms BABT
Tripathi,G; Kachhwaha,N.; Dabi,I.; Bandooni,N..
The effects of varying temperatures (12 - 44° C) on the specific activity of cytoplasmic malate dehydrogenase ((cMDH), mitochondrial malate dehydrogenase (mMDH) and lactate dehydrogenase (LDH) of some earthworms (Metaphire posthuma, Perionyx sansibaricus and Lampito mauritii) were studied. The effects of different temperatures on supernatant and mitochondrial protein contents were also investigated. The specific activities of cMDH, mMDH and LDH of the earthworms decreased gradually as a function of increasing temperature from 12 to 44°C. Higher metabolic energy was needed to maintain the activity at low temperatures. Hence, the earthworms showed increased enzyme specific activity at low temperatures. However, the protein content increased upto 28°C....

Testing Protein Digestibility in Red Grouper Epinephelus Morio using In Vitro and In Vivo Methods ArchiMer
Silva, A. F.; Escalante, Karla; Alvarez-gonzalez, C. A.; Guerrero-olazaran, M.; Cuzon, Gerard; Gaxiola, Gabriela.
The digestibility of pre-selected ingredients was evaluated for Epinephelus morio using pH-stat and multi-enzyme extract (stomach, intestine, and ceca). The degree of hydrolysis (DH) was tested with acid and alkaline proteases using crustacean meals, squid meal, fish protein concentrate (FPC), fish meal, dried whey, as well as canola paste (w/or w/o phytase), soybean meal (w/or w/o phytase), soyprotein concentrate (SPC), and wheat gluten. SPC produced DH values between 3.6-4.1%, obtained from the three different parts of the digestive tract. Canola paste produced high DH in the stomach by adding phytase to 0.8 and 2% feed. This process was repeated under alkaline conditions for DH, with 0.6-2.6% in the pyloric ceca, and 1-2.2% in the intestine. The DH from...

The Effect of Olive Cake, with or Without Enzymes Supplementation, on Growth Performance, Carcass Characteristics, Lymphoid Organs and Lipid Metabolism of Broiler Chickens Rev. Bras. Ciênc. Avic.
Al-Harthi,MA.
ABSTRACT An experiment was carried out to investigate the effect of using olive cake (OC) in broiler diets, when it is supplemented with multi-enzymes or phytase enzyme. The OC was included in isocaloric, isonitorgneous diets at 5 and 10% levels and fed to broilers from 1-28 days of age. Experimental diets were fed with or without either of the two enzymes: galzym or phytase. This resulted in 3 OC levels (0, 5, 10%) × 3 enzyme supplementations (no enzyme, galzym enzyme, phytase enzyme). This included nine treatments, and each treatment was replicated eight times with seven broiler chickens each. Feed intake, feed conversion ratio, body weight gain, survival rate, dressing, inner and immune organ´s weights (compared to live body weight); and blood lipids...

Thermotolerant fermenting yeasts for simultaneous saccharification fermentation of lignocellulosic biomass Electron. J. Biotechnol.
Choudhary,Jairam; Singh,Surender; Nain,Lata.
Lignocellulosic biomass is the most abundant renewable source of energy that has been widely explored as second-generation biofuel feedstock. Despite more than four decades of research, the process of ethanol production from lignocellulosic (LC) biomass remains economically unfeasible. This is due to the high cost of enzymes, end-product inhibition of enzymes, and the need for cost-intensive inputs associated with a separate hydrolysis and fermentation (SHF) process. Thermotolerant yeast strains that can undergo fermentation at temperatures above 40°C are suitable alternatives for developing the simultaneous saccharification and fermentation (SSF) process to overcome the limitations of SHF. This review describes the various approaches to screen and develop...

Two related thrombin-like enzymes present in Bothrops atrox venom BJMBR
Petretski,J.H.; Kanashiro,M.; Silva,C.P.; Alves,E.W.; Kipnis,T.L..
This article describes the presence of two new forms of a thrombin-like enzyme, both with apparent molecular masses of 38 kDa, in Bothrops atrox venom. Both share the ability to cleave fibrinogen into fibrin and to digest casein. Both present identical Km on the substrate BApNA. Their N-terminal amino acid sequences are identical for 26 residues, sharing 80% homology with batroxobin and flavoxobin. Two groups of monoclonal antibodies (mAbs) raised against the purified enzyme forms recognized different epitopes of the putative corresponding enzymes present in B. atrox crude venom. On Western blotting analysis of B. atrox crude venom, mAbs 5DB2C8, 5AA10 and 5CF11, but not mAbs 6CC5 and 6AD2-G5, revealed two or more protein bands ranging from 25 to 38 kDa. By...
